Scientific EMU: Aquisition of Mass Spectrometer Triple-quadrupole for Quantitative and Qualitative determination of Metabolites.

Abstract

This proposal aims to acquire a high-performance mass spectrometer of the TRIPLE-QUADRUPOLE type, model Xevo TQ Absolute, coupled to the Acquity UPLC I Class Plus chromatograph from Waters, for the ESALQ Multi-User Proteomics, Metabolomics and Lipidomics Laboratory, which comprises also the Cancer Proteomics and Metabolomics Center, an analytical unit of the USP Comprehensive Cancer Research Network C2PO, for the implementation of Omics technologies in clinical activities and cancer studies. The acquisition of this new mass spectrometer is due to the aging and obsolescence of the Q-TOF Ultima Waters equipment (PITE/FAPESP 2001/11080-8) acquired in 2004, Maldi-Tof-Tof 5800 from Sciex and Synapt G2 HDMS from Waters , both granted in the 2009 Fapesp Multiusers call (Proc. 2009/54037-7), acquired in 2011. The Q-TOF Ultima was the first equipment to become obsolete, according to a letter from the Waters company. Subsequently, Sciex's Maldi-Tof-Tof 5800, which we used for both proteomics, metabolomics and lipidomics, also became obsolete, according to a letter from Sciex. Currently, the only LC/MS we have for metabolomics and lipidomics analysis is the Synapt G2 HDMS, which is also entering obsolescence, as we were recently informed by Waters. Therefore, the acquisition of a new LC/MS with the possibility of developing quantitative analyzes will be an excellent technological upgrade for the ESALQ Multi-User Laboratory. (AU)
