Effects of the incorporation of Biosilicate doped with titanium dioxide on the efficacy, enamel mineral content and biocompatibility of experimental bleaching gels irradiated with violet LED

Abstract

Due to the adverse effects of tooth whitening on the enamel surface and pulp tissue, reducing the concentration of hydrogen peroxide (HP) and gels containing remineralizing agents become necessary. Therefore, the objective of this study will be to evaluate the efficacy, cytotoxicity and effects on the enamel surface caused by in-office whitening with experimental HP6% gels incorporated with Biosilicate¿ (BS) doped with titanium dioxide (TiO2) and irradiated with violet LED. In phase I, the BS will be doped with TiO2 and characterized regarding composition, morphology, and bioactivity . In phase II, bovine enamel-dentin discs will be randomly distributed into groups: 1) HP6%, 2) HP6%+LED, 3) HP6%+BS, 4) HP6%+BS+LED, 5) HP6%+TiO2 , 6) HP6%+TiO2+LED, 7) HP6%+BS-TiO2, 8) HP6%+BS-TiO2+LED, 9) HP35%, 10) HP35%+LED, 11) CT (control - no treatment). The whitening efficacy (¿E00 and ¿WID), surface microhardness (KHN and % Recovery - RDS), surface roughness (Ra), proportion between carbonate and phosphate, and surface morphology of the enamel will be evaluated. Experimental gels will be evaluated in terms of HP decomposition rate and presence of titanium (Ti). In phase III, enamel-dentin discs will be attached to artificial pulp chambers, and they will be whitened using the most promising protocols identified in the previous phase. The collected extracts will be evaluated for their effect on the metabolism, death oxidative stress, interleukins, glutathione, and morphology of human pulpal cells (hDPCs). Appropriate statistical tests will be conducted after exploratory data analysis (¿ = 5%). (AU)
