Abstract
Mitochondria rely on respiratory complexes and supercomplexes (SCs) to generate usable cellular energy and to maintain their shape. The alternative oxidase (AOX), found in tunicates but naturally absent in vertebrates and insects, mitigates mitochondrial dysfunction, potentially altering mitochondrial structure and function. Using diverse cells and animal models, we will integrate advanced microscopy and biochemical techniques to study the impact of AOX on mitochondrial ultrastructure and SC organization. Our findings will enhance understanding of mitochondrial adaptability, with implications for AOX-based therapies for mitochondrial diseases, ensuring effectiveness while minimizing side effects. This pioneering study bridges mitochondrial biology, evolution, and biomedical applications. (AU)
