Crystallisation and protein crystallographic studies of a number of biologically important proteins.

Abstract

The processes of life depend fundamentally on the interactions and geometrical three-dimensional (3D) structure of the molecules in the living cell. To determine 3D structures the only general method available is X-ray diffraction. The project aims to determine the high resolution crystal structures of several biologically important enzymes such as lysine-ketoglutarate reductase-saccacharopine dehydrogenase, opaque2, glucosamine-6-phosphate synthase, beta-lactoglobulin, desulforedoxin and investigate their structure-function relationships. The proteins will be crystallized using random search method as well as standard techniques of variation of precipitant concentration over a wide range of pH's and their structures will be resolved with use of synchrotron radiation.
