The purpose of this study to investigate whether stromal cells present in bone marrow (myofibrobl1asts) of patients with myeloid leukemia (AML) and myelodysplatic syndrome (MDS) have different patterns of gene expression when compared to stromal cells of bone marrow from normal donors. Preliminary studies of our group suggested that patient's cells show different growth characteristics as compared to cells obtained from normal persons. The comparative analysis of the genetic profile of the cells obtained from the two types of bone marrow will be analysed by two techniques: differential display and gene array. The molecular characteristics of stromal intercellular communication and adhesion will be studied through the determination of conexins, integrins and extracellular matrix components. In a second step normal hematopoietic cells of the umbilical cord will be cultured with either normal fibroblasts or those obtained from patients bone marrow and the profile of genes expressed will be analysed by the above techniques. Modifications on the capacity of stromal cells in affecting the pattern of proliferation and differentiation of normal hematopoietic cells will be analysed as well the possible associations with prognosis.
