In vitro fertilization in bovines

Abstract

The present project aims to master completely the stages for obtaining viable calves obtained by "in vitro” fertilization. The results from oocytes taken from slaughtered cows will be compared with those gathered by biopsy and/or aspiration in live cows whether superovulating or not. The means will be tested for maturation, preparation of the semen for "in vitro” capacitation and type of selection for motility (washing x swimming up) as well as an attempt to sex the spermatozoid by gradient. The "in vitro” fertilization, obtained in a CO2 incubator, will be the object of evaluation of the influencing factors, such as concentration of the semen, activators of the motility, individual factor of the bull and others. The development and cultivation of the fertilized structures will be conducted "in vivo” using rabbits and sheep as intermediary receptors and totally "in vitro”, using co-culture of oviducts cells in mono-layers or conditioned medium. The transfer of the blastocysts will be surgical and non-surgical, comparatively. The embryos will be sexed by H-Y antigen and karyotyping. (AU)
