Spatial analisys ot dengue transmission and molecular characterization of the virus in São José do Rio Preto, São Paulo State

Abstract

Flavivirus are responsible for a great number of severe viral diseases, which are a public health problem. Dengue and yellow fever are diseases with social and economic impact caused by that genus. The monitoring of viral circulation is important when determining the severity and magnitude of an outbreak and, associated with spatial analysis, it is an important tool for surveillance and control. This study aims at analyzing spatial patterns of dengue virus transmission and to characterize the virus molecularly through positive samples of blood, infected mosquitoes captured in residences and eggs from ovitraps. The study will be performed in São José do Rio Preto, northwest region of São Paulo state, which possesses an endemic transmission of the disease. The municipality will be divided in three different groupings according to socioeconomic level. Patients, who present fever, arthralgia, headache, or myalgia, are going to have their blood collected and will have viral RNA extracted. Afterwards, a RT-PCR will be performed for Dengue (1 to 4) and yellow fever. The positive samples will be sequenced in NS5 region. Adult mosquitoes will be captured with manual aspirators, conditioned and taken to the laboratory for identification and extraction of viral RNA. Ovitraps will be placed in outdoor sites of houses at the study region and the eggs will be taken to the laboratory to hatch. The larvae will be analyzed to check the presence of transovarian transmission of the virus. The addresses of human cases, positive mosquitoes and eggs will be kept in a data bank and the spatial analyses will be performed with estimation of density (Kernel), which analyzes the dot pattern and estimates the dot intensity of the process in all region of study. The sequences of NS5 will be compared with GenBank database and the alignment of nucleotide sequences, phylogenetic analysis and the comparison among the periods of the study will be made. (AU)
