X-Chromosomal polymorphisms study in the Brazilian population

Abstract

The human identification through DNA analysis is done by the study of polymorphic regions, such as short tandem repeats (STR). For the use of these markers in human identification, it is important to know the allelic distribution in the population to which the individual is inserted, since it can vary among different populations. It became still more relevant in X-chromosomal analysis by the fact that women have two chromosomes and men just one, that is supposed to produce larger genetic distances among populations. Ahead of the displayed one, this project has, as the main objective, to determine allelic frequencies and forensic and paternity statistics parameters for 10 X-chromosomal STRs in the Brazilian population from States of São Paulo, Rio de Janeiro, Espírito Santo and Minas Gerais. For this purpose, there will be analyzed 1000 nonrelated individuals and the DNA extraction will be done with FTA purification reagent (Whatman), followed by amplification in a multiplex non commercial system. The genotyping will be obtained through electrophoresis in automatic sequencer 3130 (Applied Biosystems) and statistical parameters will be calculated using Arlequin and Excel softwares. MEGA software will be used to construct genetic distances graphics. Along with this, this project also proposes to project and to implement a Brazilian X-chromosomal allelic frequency genetic database, available on the internet, which will compile information about X-chromosomal allelic frequencies, obtained with the results of this project, and others, published by other Brazilian researchers, working as one more available tool for the Police and DNA labs in solving forensic cases in the Brazilian context. Finally, will be analysed samples from Sao Paulo State Criminalistcs Institute and Project "Caminho de Volta" from Faculty of Medicine, University of São Paulo -USP of cases that haven't been solved yet. For this, it will be applied the results of the allelic frequencies obtained in this study. (AU)
