Abstract

The aim of this project is to assess the real biodiversity of foraminifera in the Flamengo Inlet (Ubatuba, SP) using biomolecular tools and by the relationship between the richness and diversity of the found species by molecular analysis with environmental factors sampled. This project intents to compare the biomolecular results with the recent foraminifera assemblages determined by the classical technique, which are widely used in studies of distribution of recent and sub-recent foraminifera and as proxies in environmental studies. The bottom sediment of Flamengo Inlet will be sampled using a Van Veen grab in 33 different stations to determine the foraminifera fauna and its assemblages and their relations between some biotic and abiotic factors. The material analyzed by the classical method will determine abundance, richness, and some ecological indexes for the living foraminifera fauna (Rose Bengal stained) and dead (empty tests). From the 33 stations sampled six stations will be analyzed by the molecular biology techniques which by the extraction of total DNA from the bottom sediment, realization of PCR analyze with foraminiferal-specific primers to the construction of genomic libraries for the sequencing. The calculation of richness diversity of foraminifera will be calculated by the results of these analyses. Analysis of simple correlation between the data obtained by the two different approaches (classical and biomolecular) will be made with values of richness and ecological indexes and to verify the applicability of molecular biology in studies of distribution of marine foraminifera. Multivariate analysis as PCA and MDS will be undertaken with the results obtained by the two techniques will also be used as comparative parameter. (AU)
