Abstract
The integrity of the spermatic DNA is extremely important for effective production of healthy offspring. Several techniques provide access to the integrity of the chromatin, allowing the evaluation of possible risk of impaired fertility or problems in the embryonic and fetal development. The general objectives of this research project are to test the hypothesis that the capacity and quality of packing of chromatin, its integrity and the damage caused by free radicals are different in a group of young Nelore bulls, adult and senile. To get these answers 70 Nelore bulls will be assessed, with 18 to 156 months, divided into seven groups classified by age. The thawed semen will be evaluated on: the physical parameters (motility, vigor, morphology), acrossome and plasma membrane integrity; histones substitution by protamines measured with CMA3 stainning; SCSA® - Sperm Chromatin Structure Analyses and TUNEL method; nucleotide oxidization by the concentration of 8-oxoguanine.
