Analyze of SNP rs9939609 in FTO gene in obese children

Abstract

The involvement of genetic factor in the etiology of obesity has been observed in several studies. Genome Wide Association Study (GWAS) correlated the fat mass and obesity associated gene (FTO) to a greater overall adiposity. The FTO is located on chromosome 16q12.2, encloses 9 exons and is expressed in adipose tissue, pancreatic ² cells and mainly in the hypothalamus, suggesting control of food intake and body metabolism. The variant rs9939609 (T> A) in FTO is the most studied, and is strongly associated with BMI, waist / hip circumference and weight in Caucasians. This association in South Asians is not well established, and in Chinese and Oceania populations is divergent. High-fat diets and sedentary lifestyle can intensify the susceptibility of the rs9939609 variant with obesity. The objective of this study is to assess the association between the FTO variant rs9939609 with fat, binge eating and metabolic factors in 250 pediatric and adolescent patients of both sexes (percentile> 95 for index BMI / age). There will be a control group of 50 individuals with normal weight. Patients will be genotyped by the TaqMan assay (assay ID c_30090620_10; Applied Biosystems). The fluorescence of the test will be viewed through PCR system in real time. The reliability of the genotyping will be verified by sequencing of the fragment with the rs9939609 variant of 3 individuals with different genotypes (homozygous without the polymorphism, heterozygous and homozygous for the polymorphism).
