Abstract

The main of this project is the development of an effective method to identify the Paracoccidioides Brasiliensis fungus, which causes paracoccidioidomycosis. This disease affects mainly the lungs and may be spread by hematogenic via. The most difficult part of treating this disease is the correct diagnosis, because there is a variety of fungi types and also the symptoms similarity with other diseases, such as those produced by tuberculosis. In fact, the correct identification of a particular fungus type is still difficult and expensive process, even with the recent equipments. In this context, nanotechnology brings new forms of analysis, quick and accurate in the identification of microorganisms by means of a specific gene sequence combined with metallic nanoparticles. This technology which is more current in terms of diseases diagnosis and, it is a method to identify the presence of a gene or protein fragment, with precision and a minimal amount of sample. Gold nanoparticles are functionalized with a specific genetic sequence of the Paracoccidioides Brasiliensis fungus, building a DNA nanoprobe. The fungus DNA will be extracted and amplified, and then will be used to identify the presence of the fungus by optical spectroscopy. The domain of correct fungus identification will lead the application of this analysis in body fluids that may be contaminated with microorganism. The development of the project can move to build a diagnostic system using the Raman Effect and this biosensor. (AU)
