Impact of chronic intake of caffeine associated or not with osteoporosis on the ligature induced-bone loss, bone density and healing in rats

Abstract

The aims of this project will be to evaluate the effects of the chronic intake of high doses of caffeine associate or not to osteoporosis on ligature-induced periodontitis, on the early stages of bone healing, on bone density and on bone morphological aspects in rats. Sixty Wistar female rats were assigned to one of the following groups: Control (n = 15): animals without caffeine ingestion and submitted to sham ovariectomy; Caffeine (n = 15): ingestion of 10 mg/100 g body weight/day of caffeine via drinking water for 65 days and sham surgery; Osteoporosis (n = 15): ovariectomized animals without caffeine ingestion; Caffeine + osteoporosis (n = 15): ingestion of 10 mg/100 g body weight/day of caffeine via drinking water for 65 days and ovariectomy. Fourteen days after the beginning of caffeine intake, the animals will be submitted to ovariectomy or sham surgery. Thirty five days after the beginning of caffeine intake and 21 days after ovariectomy, the first right mandibular molar will receive a ligature, whereas the contralateral molar will leave unligated. Fifty seven days after the beginning to caffeine intake and 43 days after the ovariectomy, a critical-size surgical defect (3 mm) will be created in the right tibia, while the contralateral tibia will leave without defect. Eight days after the creation of the surgical defect, the animals will be euthanized and the specimens will be processed in order to obtain decalcified sections. The area of inter-radicular bone loss (mm²), the area of new bone formation in the right tibia (mm²) and the bone density in the left tibia (%) will be evaluated histometrically. Some histological sections will be randomly selected to immunohistochemisty analysis of the osteoclastogenesis related-factors: osteoprotegerin (OPG), receptor activator of NF-kb ligand (RANKL) and tartrate resistant acid phosphatase (TRAP). In addition, the morphological aspects of the femurs will be analyzed for scanning electron microscopy. The results will be submitted to normality test to selected parametric (ANOVA) or non-parametric tests (Kruskal Wallis). The significance level for all analyses will be established at 5% (p < 0.05).
