Genetic variants envolved in folate metabolism: impact on head and neck carcinogenesis

Abstract

Changes in folate metabolism may contribute to the process of carcinogenesis by influencing the reactions of DNA methylation and genomic stability. Polymorphisms in genes encoding enzymes involved in the folate pathway may alter enzyme activity and consequently interfere in concentrations of homocysteine, S-adenosylmethionine and other products of metabolism which are important for DNA synthesis and cellular methylation reactions. The objective of this project is to investigate polymorphisms of BHMT, DHFR, SHMT, MTHFD1 and TC2 genes, which participate in the metabolic pathway of folate, in the risk for head and neck cancer and also investigate the association between these polymorphisms and the clinical characteristics of patients. The genetic changes will be investigated by the techniques of Polymerase Chain Reaction (PCR), Real-Time PCR and analysis of Restriction Fragment Length Polymorphism (PCR-RFLP). The results of this project will contribute to understanding the involvement of genes from the folate pathway in the development of squamous cell carcinoma of head and neck and to clarify the mechanisms leading to the tumorigenesis process.
