Effects of thymulin 5cH in BCG-induced granuloma: the B-1 cell participation

Abstract

In previous studies, it was observed that thymulin prepared homeopathically (5cH potency) is able to improve broiler production, as well as to modulate the Ehrlich tumor development and BCG induced-granuloma in mice, involving immune modulation mechanisms. Herein, a deeper approach about these modulation mechanisms is proposed, with special focus on the leukocyte migration to the lesion, mainly B-1 cells. Male Balb/c mice will be inoculated with BCG, treated orally with thymulin 5cH or vehicle, and the subcutaneous granuloma and the local lymph node will be studied, regarding to the phenotypical characterization of inflammatory cells using immunohistochemistry and flow cytometry techniques. In the first case, the number and distribution of positive cells for CD3, CD11b, CD54R, pan-cytokeratin (epithelioid cells) will be evaluated. In the second case, the quantification of positive cells for CD11b, CD19, CD23, CD5, CD25 (regulatory T cells), CD4, CD8 and CD11c (dendritic cells) will be done. The statistical analysis will be Student "t" test or X2, being p=0,05. (AU)
