Abstract

The genome project of X. fastidiosa will bring up the total DNA nucleotide sequence however the expressed sequences will be not known. To better understanding of the panel of the transcripts by the genome it would be necessary the analysis of the transcripts expressed in certain stages of the bacterium development. The most important stages in a prokaryotic and pathogenic bacterium are the growth phase and the interaction with the host. The present study intends to analyze in first instance the presence of induced mRNAs during the cultivation in a culture medium and in a second stage, the total pool will be compared to the mRNAs, which are specifically induced by citrus extracts. The APES (arbitrarily primed expressed sequences) approach will be applied. This strategy, named APES by our laboratory, will search for total mRNAs, conversion to cDNA using arbitrary primers, cloning and sequencing of the flanking ends. The terminal sequencing will be compared to the Genome sequence of X. fastidiosa to search for the corresponding genes and functional assignment. (AU)
