Abstract

The purpose of this project is to establish conditions for gene disruption in Xylella fastidiosa, aiming at genes potentially involved in bacterial pathogenesis. Two main strategies will be attempted: (a) electroporation of Xylella fastidiosa with a suicide vector containing the gene of interest interrupted with a cassette encoding an antibiotic resistance gene; (b) conjugation of Xylella fastidiosa with an Escherichia coli donor strain carrying the suicide vector described in (a). Once the Xylella fastidiosa strains carrying a disruption in each of the target genes have been obtained, pathogenicity tests will have to be carried out. (AU)
