Abstract
This proposal has the objective of adapt a genetic transformation protocol, for commercially important varieties in Brazilian citrus industry, with emphasis to sweet orange. This protocol could be further used to introduce genes identified in the Function Genomics Program (FAPESP). Genetic transformation will be performed via Agrobacterium, with EHA 105, LBA 4404 and GV 2260 isolates, with kanamycin gene for selection and GUS as gene reporter. Different explants will be evaluated for transformation efficiency, such as epicotyl segments from in vitro germinated seedlings, intermodal segments from seedlings germinated in a greenhouse, and mature buds, from budded trees in a greenhouse. Embryogenic calli will also be used as an explant source. Regenerated plants will be evaluated by GUS test and those identified as GUS+ will be analyzed by RAPD for confirmation of genetic transformation. (AU)
