Abstract

This proposal is devoted to the development of methods to assess for metal contamination in fish, mainly related to tilapias (Oreochromis niloticus). By using fish bioassays, the lethal concentrations (CL50) for Cd, Cu and Cu+Cd and the respective metalothioneins (MTs) formation, as a detoxification mechanism, will be investigated. The formation and elimination of metalothioneins in organs like liver and gills will be the aim of the analysis, which will be supported by the stable isotopes 111Cd and 65Cu.It is also described the analytical protocols related to sample preparation and isoform separation by capillar electrophoresis towards detection by mass spectrometry ICP-MS. The experimental is designed in blocks with repetitions, being the data analysed by variance analysis (ANOVA) and regression. (AU)
