Evaluation on the interaction between two osteogenic proteins and different bone grafts in the bone healing process of calvarial tissue in Wistar rats

Abstract

With the discovery of proteins with osteogenic potential, it was radically changed the thinking about the bone induction, these proteins are multifunctional and with wide effects on cell growth and differentiation in many organisms. The role of BMPs in critical bone defects has been studied in the literature, but it has not been examined in large extension. This study aims to evaluate the bone healing process in critical bone defects performed in rat skulls, using different types of bone grafts, the osteoinductive protein rhBMP-2, and the association between these grafts and this protein, by histomorphometric, immunohistochemical methods, and statistical analysis. It will be used 112 Wistar rats, male (250 g) to be divided into 8 groups, and a period of time until the sacrifice of 4 and 6 weeks. The groups are the following: 1) particulate autologous bone graft; 2) particulate allografts, 3) heterologous bone graft (lyophilized bovine bone), 4) particulate autologous bone graft plus 5 µg rhBMP-2, 5) particulate allografts plus 5 µg rhBMP-2, 6) heterologous bone graft (lyophilized bovine bone) plus 5 µg rhBMP-2, 7) 5 µg rhBMP-2, 8) only critical bone defect. The results for new bone formation will be evaluated by histomorphometric and immunohistochemical methods, and the resulting data from these assessments will be submitted to statistical analyses.
