Functional analysis of Pou5f1 (Oct4) gene marker of stem/progenitor cells in cultured tumor pituitary cell lines

Abstract

The maintenance of tissue in the adult organism requires a constant supply of new cells to replace cells lost to stress, injury or natural death. The cell replacement is possible through the activity attributed to tissue-specific stem cells. The anterior pituitary is an important regulatory role in the metabolism of the adult organism by regulating the hormone production. The cellular composition of the gland undergoes changes in response to hormonal signals, for example, the increase of the gland during puberty and pregnancy. Besides the known role of stem cells during embryogenesis pituitary and its maintenance in adulthood. Another emerging concept of stem cell biology is its connection with carcinogenesis, underscored by the consistent presence of a compartment of undifferentiated cells connected with the initiation, growth and maintenance of tumors such as breast, glioblastomas, among others. It was recently shown in humans that ACTH-producing tumors have an increased expression of the gene Pou5f1 (Oct4), compared to normal pituitary gland. This gene is expressed in embryonic stem cells and stem / progenitor cells of many tumors. Therefore, the objective of this study is to evaluate the role of gene Pou5f1 (Oct4) in proliferation and hormonal phenotype of tumor cells from pituitary cell cultures of mouse and rat. (AU)
