Identification and characterization of physiological races of Fusarium oxysporum F. sp. phaseoli in common beans

Abstract

Fusarium wilt is an important common bean disease that causes great losses in production as the majority of the cultivars are still susceptible and the fungus is easily disseminated. In literature, two races are reported in Brazil; however, in more recent studies, divergent results have appeared, suggesting that new races have arisen. Based in this context, the objective of the present work is to monitor the infection of Fusarium oxysporum F. sp. phaseoli through real time PCR in differential varieties such as 'BAT-477', 'A-211', 'Mortiño', 'MFS465-63-1', 'Frijolica', 'RIZ 30', 'Calima', 'TIB 3042', 'IPA-1' genotypes (proposed by CIAT), and 'Preto Uberabinha', 'BBL-274', 'Tenderette' and 'IPA-6' as proposed by Nascimento (1992). Another goal is to identify resistance sources that present carioca and black tegument types, making it easier to incorporate genetic resistance. For this purpose, 300 accesses of the IAC germplasm bank will be inoculated with three different isolates; one of them being a Brazilian race, one of the American race and another one of a non-defined race (NDR). In an attempt to characterize the interaction of the fungus in susceptible and resistant host tissues, three enriched microsatellite libraries will be developed for Brazilian, American and NDR races of Fusarium oxysporum F. sp. phaseoli so that specific microsatellites can be developed and the genetic diversity of the isolates can be characterized.
