Osmotic stress in the control of direct somatic embryogenesis em genotypes of Coffea arabica

Abstract

The coffee market has high demand for hybrids of Coffea arabica L. that have good quality drink. However, the multiplication of these hybrids must be vegetatively because plants from germination of seeds lose the genetic characteristics of the hybrid, due to genetic segregation. Somatic embryogenesis (ES) allows the vegetative propagation of these hybrids. This process occurs in two ways, indirect somatic embryogenesis (ESI), consisting of the stages of embryogenesis and organogenesis, somatic embryogenesis or direct (ESD) at one stage. The comparison between the two approaches indicates that the ESD is more advantageous than ESI, since the embryos are formed with reduced inputs, labor and time. However, genotypes of C. arabica not respond efficiently to ESD, this means they have no or low production of somatic embryos are formed and the long period of time. Thus, there is a need for more efficient implementation of ESD in genotypes of arabica coffee. Stressors such as osmotic has been found to increase the capacity of ES in different plant species. Thus, it is intended to define the effect of osmotic stress in controlling the occurrence of the ESD in three hybrids of C. arabica, with good sensory quality, and cv Mundo Novo, in order to increase the production of somatic embryos, reduce your training time, develop a more efficient protocol for the implementation of ESD in this species and the production of seedlings of selected hybrids that will later be subjected to the condition field for evaluation of agronomic performance. For this, we performed six experiments, five with the induction of osmotic stress in leaf explants and on the evaluation of regenerated seedlings under this condition. (AU)
