Abstract

The resorption of the mineralized tissues is one of the great preoccupations of the Dentistry, because the control or interruption of this process is not always achieved in the clinic by the lack of knowledge of their immunopathogenic mechanisms. The present research project aims to characterize the cellular and molecular mechanisms involved in the resorption of mineralized tissues (tooth and alveolar bone), through the experimental induction of periapical lesions in wild-type mice, and assess the specific role of intercellular adhesion molecule-1 (ICAM-1) in this process using transgenic (knockout) mice. To induce resorption of apical and periapical tissues, will be held coronal opening of the 1st molars and exposure of root canals to oral environment for contamination. After the death of the animals samples, taken at different experimental periods, will be evaluated by conventional and fluorescence microscopy (morphometry), histoenzimology (TRAP), immunohistochemistry, immunofluorescence and Real-Time PCR to assess cytokine expression (IL-1alpha, TNF-alpha, IFN-gamma, IL-4, IL-6, IL-10 e IL-17) and factors involved in osteoclastogenesis (RANK, RANKL, OPG e catepsin K). Potential immunopathological findings could provide basis for better understanding of the mechanisms involved on resorption of mineralized tissues, as well as generate data with potential applicability in the development of new tools for early diagnosis of pathological bone resorption, or new therapeutic targets.
