Effects of Tityus bahiensis venom on the maternal reproductive development and the participation of cytokines and of growth factors on the changes observed in the offspring of dames treated with the venom on the perinatal period

Abstract

In previous papers, we have observed that, when the Tityus bahiensis scorpion venom is administrated to dames during the 10th or the 16th pregnancy days, some changes occur on the physical and reflexological development of the offspring during the perinatal period, as well as some behavioral changes on the offspring when they reach adulthood. Reports throughout the literature have demonstrated that growth factors interfere in the embryo development and cytokines may be related to morphological and behavioral alterations. Bearing this in mind, our aim is to investigate whether the changes found in the offspring may be due to the venom's interference over these mediators when administrated to the dames. Therefore, we will evaluate the levels of IL-1, IL-6, IL-10, INF-³ and TNF-± and the levels of NGF (neuronal growth factor), and BDNF (brain derived neurotrofic factor) by means of an enzyme immunoassay and the NFºB (nuclear factor kappa B) expression in shift-gel assays for embryos with 10 or 16 days and for the brain of new-born pups from dames treated with the venom on the 10th or 16th gestational day or for brain of new-born pups from dames treated with the venom on the 2th, 10th or 16th day of lactation. In addition, we will also ascertain whether the venom administration causes changes to the maternal reproductive development and the fetuses' morphology. The pregnant dames will be evaluated as to weight gain during gestation, and, on the twentieth gestational day, they will be submitted to laparotomy. The uterine horns will be exposed, and the pups will be taken out, together with their placentas. Pups and placentas will be counted, weighted and externally analyzed as to their general appearance, sex and vitality. Then, each litter will be randomly divided into two different groups, in order to analyze possible visceral or skeletal malformations. In the uterus, we will check for the presence of reabsorptions and implant points. The corpora lutea present in each of the ovaries will be counted, as to evaluate the amount of eggs that were eliminated. (AU)
