Porous titanium and biomimetic processing: effects on gene expression of osteoblastic cells derived from human alveolar bone

Abstract

The osseointegration depends on cell-biomaterial interation. Titanium is one of the most important biomaterials for implantation in bone tissue used in medical and dental areas. Different topographies of the implant have been studied to improve osseointegration. In addition, biomimetic surfaces that incorporate bioadhesive properties are promising for surface modification of biomaterials. The objective of this study is to investigate the behavior of human osteoblastic cells and osteogenesis in vitro on porous titanium specimens with different porosities, with and without biomimetic treatment, comparing it with dense titanium. It will be fabricated, by powdered metallurgy, disks of pure titanium grade II, divided into six groups: 1)dense titanium sample (Ti), 2)sample with 30% of porosity (30% pores of 300um of diameter), 3)samples with 40% of porosity (40%), 4)dense Ti-sample with biomimetic treatment (TiB), 5)sample with 30% of porosity with biomimetic treatment (30%B) and 6)samples with 40% of porosity with biomimetic treatment (40%B). Metallographic analysis will be made and DRX (X-ray Scattering) of the samples. In vitro data will be obtained regarding adherence, morphology and cell viability, total protein content, alkaline phosphatase activity, amount of nodular formations and matrix gene expression by RT-PCR (real-time polymerase chain reaction) of the following genes: alkaline phosphatase, prostaglandin E2, ²1 integrin, collagen 1, osteocalcin, osteopontin, osteonectin, bone sialoprotein, TGF-², osterix, Runx2, Interleukin 6, colony stimulating factor macrophage - MCS-F and apolipoprotein E. Data will be evaluated by statistical tests ANOVA test, Tukey's multiple comparison, Student's t test and Kruskal-Wallis test, all at 5% significant level. (AU)
