Search and identification of peptide of amelogenin isoforms from the X chromosome or Y chromosome in human teeth

Abstract

Recently we demonstrated that it is possible to identify enamel-specific peptides in human mature enamel of teeth contemporary teeth and also from 1000-years mummy teeth. Among the enamel-specific peptides identified, one amelogenin peptide was identified, which is very interesting because the sequence of this peptide is encoded in the x-chromosome specific sequence. These results suggest that it shall be possible to further refine our current techniques and search for peptides that are specific for either the X-amelogenin isoform or the Y-amelogenin isoform. Up to now such specifi peptide sequences have not been identified from mature enamel samples, but these sequences are well known from the peptide sequences (and cDNA knowledge) found in immature enamel. This study aims at identifying X-amelogenin or Y-amelogenin specific peptides in recently extracted human teeth and in ancient teeth (mummy teeth) that belong to either man or woman according to gender idenfication methods (PCR). The results of this study will be very important for the identification of the gender of persons and has a bearing for archaeological and forensic research. (AU)
