Serum amyloid A and melanomas: receptors, matrix metalloproteinases and immune system interactions

Abstract

For some types of tumors it was evidenciated a positive correlation between serum amyloid A (SAA) production and malignance. Our group has described the role of SAA as a growth factor stimulating proliferation of different cell types. In two glioma lines, beyond the effect in proliferation, SAA modified migration and invasion and it was dependent of matrix metalloproteinases. Interestingly, for one line, SAA cause a decrease in MMPs activities and consequent migration and invasion inhibition, while for the other one occur the opposite effect. This result leads us to this project that objective evaluate the SAA effects in a group of tumor lines to define it contribution in the malignance. Melanomas were chosen for it aggressiveness, being studies for competent groups in São Paulo and for this tumor SAA was considered a prognostic factor. Will be used a group of cells with distinct patterns of mutations, cultivated both in monolayer and imunocompetent tridimensional (3D) models (collaboration with Profa. Silvya Stuchi - FCF/USP) and clinical samples (collaboration with Gilles Landman- UNIFESP). Preliminary results with two melanoma lines showed a different profile in gene expression of SAA, migration and invasion. Another important question is the definition of SAA receptors in tumor cells. In this sense, also in a preliminary study, we identified that the expression of Toll-like receptor (one of the SAA receptors) is higher in the more responsive lineage. (AU)
