Abstract

Phrynops geoffroanus will be captured monthly at Piracicamirim and Piracicaba rivers during the whole visit period with nylon fishing nets.Blood samples (5 ml) will collected from at least five adult male and five adult female turtles (up to ten samples from each sex will be collected if available) in every month of the year for one year. The blood will be held on ice until red cells are separated by centrifugation (an aliquot of whole blood, 100 ml will be set aside for any potential genetic analysis). The plasma will be divided into two separate tubes and frozen until ready for assay. The following steroids (testosterone, estradiol, progesterone, and corticostereone) will be measured in the plasma samples using radioimmunoassay (RIA) as described in the references below, or using the non-radioactive enzyme-linked immunoassay (ELISA), if available. If time permits, plasma calcium and magnesium will be assayed using atomic absorption spectrophotometry (AA), and analysis of plasma lipids using standard clinical techniques will be undertaken. In addition, analysis of whole blood for heavy metal contamination will be tested using AA or a similar technique. (AU)
