Abstract
The understanding of molecular structure and protein function mechanisms is one of the most interesting challenges in Science due to the crucial role that these molecules play in all biological processes for life appearing and maintenance. In this way, elucidating the molecular mechanisms that can interfere on protein and peptide activities as a function as the changes promoted by external agents and specific bindings is essential to natural processes comprehension and for biomaterials development as biofunctionalized scaffolds, biosensors and biofuel cells. From these studies there are several spectroscopic methods that allowed not only the characterization of structural changes, but also the thermodynamic parameters determination for the biomolecules that are being studied. From the several physic-chemical known methods, steady-state fluorescence spectroscopy has been widely used at most biophysics and biomaterials studies by reason of the high sensitivity for studying changes at fluorophors vicinities in both intrinsic (aromatic amino acids) e and extrinsic. In some cases, is even possible monitor tertiary conformational changes of proteins and peptides. Hence, it is being proposed in this project the utilization of the steady-state fluorescence spectroscopy technique to enlighten the conformational changes in different biomolecules with application to biomaterials development.
