Abstract

Several biotechnologies are being developed to promote the conservation of the genetic material of stallions with high zootechnical value, among which we highlight the collection of sperm from the epididymis of animals who have suffered some trauma or illness preventing the collection of semen, death or euthanasia. However, these sperm cells do not come into contact with seminal plasma, which is important because it contains proteins that participate in processes related to the protection and retention of sperm in the binding sites of sperm in the fallopian tube reservoir. The colonization of the oviduct sperm reservoir is an important phenomenon responsible for reducing the risk of polyspermy by releasing a small amount and sufficient sperm for fertilization at the time of ovulation and prevents premature sperm capacitation maintaining in basal levels the concentration of intracellular calcium. This study aims to evaluate the binding ability of sperm cells from the tail epididymis to oviduct cells cultured in vitro in the equine species. For this, sperm parameters are compared between sperm in natura and frozen obtained from ejaculated and tail epididymis from ten stailons, which are incubated with cells istimo from mares cultured in vitro. After the incubation period, the explants are removed from the droplets with the minimal medium, the remaining volume of the drops will drawn and the concentration of sperm remaining is counted. The number of sperm which bound will estimated by subtracting the number of spermatozoa after the incubation period of the initial concentration of spermatozoa. (AU)
