Determination of the frequency of viruliferous Brevipalpus phoenicis mites in citrus orchard and studies of vector mite and CiLV relations

Abstract

Quantifying the number of Brevipalpus phoenicis mites infected by Citrus Leprosis Virus (CiLV) may contribute to improve the mite sampling techniques and disease management. In addition, the increase in knowledge about the relation between vector mite and CiLV disease would be fundamental to adequate the leprosis control methods. The primary objective of this project will be to quantify the frequency of viruliferous and aviruliferous B. phoenicis according to the amount of leprosis inoculum. Additionally, it is intended to perform a histological and ultra-structural study of B. phoenicis internal organs to expand knowledge about the relation between vector mite and the disease. The trials will be conducted in laboratory and in the field. Firstly, sweet orange plots with leprosis symptoms will be selected. The symptomatic plants and some plants around them shall be marked by GPS device. After that, a population survey of B. phoenicis will be performed. The orange fruit will be collected and sent to the laboratory for the B. phoenicis removal. Then, an application of acaricide will be performed over the field marked plants. Lastly, monthly, after the application will be performed population survey and collection of B. phoenicis. The collected mites will be used to check infectivity of CiLV through reverse transcription polymerase chain reaction (RT-PCR) using specific primers to the virus and for the ultra-structural studies on transmission electron microscope (TEM). The results of viruliferous and aviruliferos B. phoenicis frequency will be analyzed, and also, the ultra-structural results, localization and characterization of CiLV presence in mite will be described through photomicrographs examination, which were obtained in TEM.
