Leishmanicidal activity of palladacycle complexes: in vitro and in vivo inhibitory effect of DPPE 1.1 on Leishmania (Leishmania) amazonensis and evaluation of immune responses in infected mice treated with DPPE 1.2

Abstract

Previous results showed the in vitro and in vivo activity of the palladacycle complex DPPE 1.2 against Leishmania (Leishmania) amazonensis. The treatment of L. (L.) amazonensis-infected BALB/c and C57BL/6 mice with DPPE 1.2 resulted in a significant decrease of lesion size and parasite load at concentrations that are non toxic to the host. There was an increase of CD4+ and CD8+ T lymphocyte expression in the popliteal and inguinal lymph nodes of mice treated with DPPE 1.2 followed by an increase of IFN-gamma and a reduction of TGF-² secretion. These results indicate that the treatment with DPPE 1.2 modulated the immune system in treated mice. Our previous data also demonstrated the in vitro and in vivo activity of the palladacycle complex DPPE 1.1 against L. (L.) amazonensis. However, the leishmanicidal mechanism of this compound was not studied. The goal of the present project is to explore the immune mechanisms induced after treatment of L. (L.) amazonensis-infected mice with DPPE 1.2, as well as to extend the study on the in vitro activity of DPPE 1.1 on L. (L.) amazonensis and the possible immunomodulation exerted by this compound in treated animals. (AU)
