Abstract

This study aims to evaluate the hibiscus sabdariffa supplementation effects on kidney function changes in Wistar rats with palatable hyperlipidic diet (PHD) obesity induction. 48 male Wistar rats will be allocated into four groups (n=12): control group (CG)-animals fed with commercial diet for rats and ad libitum water intake; Hibiscus control group (HCG)- animals fed with commercial diet for rats and ad libitum intake of hibiscus in aqueous solution; obese Group (OG)- animals fed with PHD and ad libitum water intake; Hibiscus obese Group (HOG)- animals fed with DPH and ad libitum intake of hibiscus in aqueous solution. Will be used dry extract of hibiscus, diluted in water in a concentration to provide anthocyanins intake of 100-200 mg/kg/day. The animals will receive the diets and hibiscus for 16 weeks. The rats will be weekly weighed, monthly evaluated for food and water intake and allocated in metabolic cage (24 hours) for urine collection and determination of urine output, total protein, creatinine, sodium, potassium, chloride and urinary calcium. A week before the end of the study will be tested for glucose tolerance. At the end of 16 weeks, the animals will be anesthetized, blood was taken for biochemical (urea, creatinine, sodium, potassium, chloride, calcium, serum triglycerides, total cholesterol, LDL and HDL) and hormonal studies (leptin and insulin). Immediately after the death, kidneys and retroperitoneal fat will be collect and weigh, then performing right kidney freezing for molecular analysis, to evaluate the kidney transcriptome expression by the transcriptoma shotgun sequencing or RNA-seq technique, and histopathology of the left kidney to evaluate glomerular membrane, fat tubular accumulation and tubulointerstitial fibrosis.
