Differential transcript profile in mesenchymal stem cells obtained from the menstrual flow (MenMSCs) of women with endometriosis

Abstract

Endometriosis, a gynecological disease that affects 6 to 10% of women of reproductive age, is considered to be a public health problem both because of its impact on the physical and psychological health of the patients, and because of the socioeconomic impact caused by the costs of its diagnosis, treatment and monitoring. Several mechanisms have been proposed to explain the origin of the disease, such as: retrograde menstruation, genetic predisposition, metaplasia of peritoneal cells, and changes in the immunological response. During the last decade, cells with properties similar to those of stem cells (SC) have been identified in the endometrium and in menstrual blood, and have been suggested to be related to the etiopathogeny of the disease. Thus far, however, the profile of gene expression (transcriptome) of these cells has not been defined. The proposal of our group is to isolate, characterize and identify the transcripts of these cells desquamated during menstruation (MenMSCs) in women with and without endometriosis, aiming to understand the possible involvement of these cells with the etiopathogenic aspects of the disease. This proposal will also provide for the institution hosting the establishment of new methodologies for the derivation and culture of a promising SC line (MenMSCs) with certain advantages over the SC types already available for research in this institution, permitting future possible applications to other research lines, thus favoring the nucleation of new research groups.
