Abstract

Introduction: The pathogenic mechanisms responsible for the reduction of fertility in women with endometriosis remain poorly understood, being questioned a possible deleterious effect of this disease on ovarian steroidogenesis, folliculogenesis and oocyte quality. As human oocytes are extremely rare and their use in invasive studies, that generally impossible their use in assisted reproduction procedures, is usually not possible, the identification of noninvasive biomarkers capable of predicting oocyte quality, is highly desirable. In this context, the Cumulus Cells (CCs) can be used as biomarkers of oocyte quality. Analyze the gene expression profile in CCs could bring valuable information about dysregulated pathways involved in the genesis of infertility related to endometriosis. Objectives: Determine the set of mRNAs expressed (transcribed profile) in CCs of infertile women with and without endometriosis (stages I/II and stages III/IV) undergoing controlled ovarian stimulation for ICSI, by next-generation sequencing; and compare the profile of transcripts between the groups proposed. Later, we propose to evaluate the expression of 3 target genes, to be selected according to their differential expression and its relevance in the pathophysiology of infertility, endometriosis and acquisition of oocyte quality, by real-time PCR. Methodology: In the study we will analyze the CCs of 30 patients, 10 controls, 10 with endometriosis I/II and 10 with endometriosis III/IV. All patients will be subjected to the EOC to perform ICSI. Immediately after oocyte retrieval, the CCs are isolated and stored for the Next-Generation Sequencing (RNA-Seq), moreover, analyzes of the expression of 3 target genes by real-time PCR will be performed.
