Abstract

Pemphigus Vulgaris has been associated to HLA alleles in different populations, mainly HLA-DRB1*04:02 and HLA-DQB1*05:03. However, these alleles are not always present in PV patients. Preliminary study of this institution has investigated the possible correlation between HLA-DR alleles and Pemphigus Vulgaris by typing a cohort of 36 patients and 162 healthy controls from a Brazilian highly admixed population. Significantly higher frequencies of HLA DRB1*04:02, DRB1*08:04, and DRB1*14 alleles were observed in the patients than in the control individuals (47.2% vs. 1.2%; 36.1% vs. 4.3%; 27.8% vs. 7.4%, respectively). This work describes for the first time a strong association between the DRB1*08:04 allele and pemphigus vulgaris in the Brazilian population. The results raise a suspicion of the DRB1*08:04 allele being involved with the disease. The present study has the purpose of doing a High-resolution typing of nearby loci DQA1 and DQB1 on the previously extracted DNA to differentiate if the association is inherent to the HLA-DRB1*08:04 allele or to a specific haplotype and also will amplify the number of patients into fifty six, by typing HLA-DR and DQ in all of them.
