Detection and characterization of Escherichia coli avian pathogenic (APEC) in domestic backyard poultry in the region of Ribeirão Preto, SP

Abstract

The creations of "domestic backyard poultry" (DBP) is the main activity performed by small producers in the world. Sanitary problems are the main limiting factor of these creations, which may contribute to the dissemination of diseases to both poultry and the consumer. Among the bacterial agents cause of diseases in birds there is the Escherichia coli pathogenic for poultry (APEC). The aim of this study is to detect APEC strains present in the creations of country chickens in the region of Ribeirão Preto - SP. Therefore, the specimens will be collected through a cloacal swab and oropharynx of GFQ. The samples will undergo a screening by PCR for the detection of APEC. Samples positive for APEC strains will be searched characterized by PCR for genes cvaC, ironN, iss, iucD, iutA, sitA, traT, tsh, fimH, papC, papG II and cnf. Additionally PCR, the isolates of APEC will be tested for antimicrobial susceptibility test to determine the serogroups and will be evaluated for pathogenicity by inoculation test in vivo in chicks from one day. Also will be analysed the profile of sensitivity of the isolates to hydroalcoholic extracts Eugenia uniflora (cherry), Cuphea carthagenensis (seven bleending), Vernonia polyanthes L. (assa fish) and Arctium minus (burdock). Furthermore, will be established epidemiological relationship of isolates by PFGE and phylogenetic analysis by PCR for ChuA, YjaA and TspE4C2. The detection of APEC and various virulence genes may evidence what genes (or set of genes) are associated with the disease, furthermore can show if the DBP can be reservoirs of potentially pathogenic bacteria to human.
