Abstract

Cadmium (Cd) difficult the development of plants, whereupon can be minimized by sulfur (S) supply to plants grown in contaminated environments and the use of tolerant genotypes with high mass production for phytoremediation. The main objective is to evaluate the effect of S on the Cd uptake and its distribution on plant tissue, evaluate the ultra-structural changes on the root and leaves tissues, assay the oxidative stress and the photosynthetic parameters [CO2 assimilation (PN), stomatal conductance (gs), transpiration (E), intercellular CO2 concentration (pI), chlorophyll fluorescence (the effective quantum efficiency of photosystem II and apparent electron transport)] driven by Cd (111Cd) uptake, as well as to determine Cd content on the apoplast and symplast of Panicum maximum cv. Tanzania. The hypothesis to be verified is that well S-nourished plants increase the synthesis of glutathione and phytochelatins, attenuating oxidative stress caused by Cd excess. It is also expected that the chlorophyll concentration and the effective quantum of photosystem II increase with the S supply due to the attenuation of oxidative stress and lower Cd transport from roots to shoots due to higher Cd apoplast uptake. Higher Cd apoplastic accumulation (than symplastic) has been described as an effective mechanism of heavy metal tolerance. Therefore, it is expected that the appropriate S supply to Panicum maximum cv. Tanzania may contribute to the advancement of phytoremediation in environments with low to moderate Cd contamination. The experiment will be carried out in a greenhouse, using nutrient solution with three S rates (0.1, 1.9 and 3.7 mmol L-1) and three Cd rates (0.0, 0.1 and 0.5 mmol L-1), in a factorial 3x3 experimental arrange. These following evaluations will be held at the end of the plant growth: morphological, productive, nutritional parameters, Cd-apoplast and symplast content, ultra-structural changes, soluble protein content, H2O2 content, lipid peroxidation, reduced glutathione, oxidized glutathione, the total glutathione, activity of superoxide dismutase, ascorbate peroxidase, catalase, guaiacol peroxidase and glutathione reductase, PN, gs, E, pI, minimal and maximal chlorophyll fluorescence, fluorescence in dynamic and maximal equilibrium state. The results will be submitted to analysis of variance and the treatment means are compared by Tukey test at 5% significance. (AU)
