Abstract

The aim of this study is to investigate the effects of the histaminergic H4 agonist VUF-8430 microinjections into the cerebellar vermis in mice subjected to the elavated plus maze (EPM), inhibitory avoidance (IA) and Rotarod test in the consolidation of emotional memory and the motor control. After surgical recovery from the procedure to implant a cannula, the animals will receive saline or VUF (0.15 nmol; 0.49 nmol; e 1.48 nmol/ 0.1 ¼l) administered after the first day of test. The Experiment 1 on EPM will be performed on two consecutive days (T1 and T2) andthe animals will be free to explore the maze for 5 min. Conventional measures will following: frequencies of open- and enclosed-arm entries, total arm entries, total time spent in the open and enclosed arms, and total time spent in the central area. Indexes on T1 will indicate the level of anxiety shown by the animals and the decreased activity in the open arms between T1 and T2, will infer the emotional memory. The Experiment 2 on IA will be performed on two consecutive days (D1 and D2). Latencies will infer the emotional memory. The Experiment 3 on the Rotarod test will be performed on two consecutive days (R1 and R2). The rod-rotation speed will be 16 rpm for 300s and the latency to animal's fall is automatically measured. In R1, the animals will be habituated for 5' and after 15' will be tested. After the end of R1, the animals will receive pharmacological treatment. In R2, the animals will be re-exposed under the same experimental conditions of R1 but without the microinjections, featuring motor learning. The participation of the histaminergic agonist H4 in the emotional memory modulation will determine another investigation, if the microinjection of VUF-8430 activates the phosphorylation of CREB transcription factor, using the Western blotting technique (Experiment 4).
