Abstract

The objective of this study is to assess the response of subcutaneous connective tissue of isogenic mice after the use of Photodynamic Therapy antimicrobial (aPDT), using two photosensitizers (phenothiazine and curcumin dyes), at different times of laser application. BALB/c isogenic mice (n = 262) will be used and divided into three experimental steps: first step (n = 89) - standardization of the concentration of the phenothiazine dye; second step (n = 126) - comparison between the dyes and standardization of time of exposure to light; third step (n= 47) - response of subcutaneous tissue contamination with bacterial LPS and application of aPDT through standardized protocols in the first and second stages. At the end of each experimental periods (7, 21 and 63 days), the connective tissue subcutaneous tissue of the area in which it was applied the aPDT will be removed and submitted to histotechnic processing routine. It will performed a qualitative description of the inflammatory process, a semi-quantitative analyses (scores) and a quantitative one (cell count). In addition, immunohistochemistry for proinflammatory mediators, neutrophils, and macrophages will be done. The numerical data will be analyzed by statistical software GraphPad Prism® 5 (GraphPad Software In., San Diego, California, USA), using the appropriate test in accordance with the nature of the data obtained.
