Integrated studies of quantitative genetics and genomics for economically important traits in tambaquis (Colossoma macropomum)

Abstract

Fish consumption is associated with the different feeding habits of human communities. The continuous demanding of fish meat is mainly due to the health benefits of this food source. Among the various factors used in aquaculture operations, genetic improvement programs is one of main drivers to the productivity growth worldwide. Likewise, livestock breeding programs have achieved considerable outcomes to improve economically important traits for animal food production. Tambaqui, Colossoma macropomum (Cuvier, 1818), is the largest representative of the Characiformes order found in the Amazon and Orinoco basins. Currently, it's among native Brazilian fish species with the higher farming production with 88.719.00 t in 2013. We propose a coordinated set of aims to study and produce information on genetic parameters of different traits through diallel cross among different phenotype individuals, as well as to use molecular strategies to support the breeding strategies used. We will study two traits for the genetic parameters assessment: absence of intramuscular bones and loin eye area (LEA) using tagged tambaqui families with ultrasound images to obtain the mentioned phenotypes after growout period. Moreover, we will carry out Genotyping by Sequence to produce a SNP panel to attempt to associate SNPs markers with phenotypes, and microsatellites polymorphism evaluation of growth hormone gene (GH). Tambaqui breeding, family production and growout will be held in the BioFish Ltd. Hatchery located in Porto Velho - RO. The molecular procedures will be carried out in the Laboratory of Aquatic Organism Genetics and Aquaculture (LAGOAA) at the University of Mogi das Cruzes with collaboration with Laboratoy of Animal Biotechnology at the University of São Paulo - Piracicaba. The long-term goal of our research is to understand the genetic and molecular parameters underlying the mechanisms that control the phenotypes proposed in the current proposal. And, to contribute to establish further breeding programs to develop new more productive strains of tambaqui, as well as to other native fish species of importance for the Brazilian aquaculture.
