Topical use of heterologous mesenchymal stem cells from canine adipose tissue in the treatment of keratoconjunctivitis sicca in dogs

Abstract

Stem cells have the tissue regeneration capacity due mainly to its paracrine action at the site of its application, and it can thus regenerate the lacrimal glands destroyed by the action of antibodies that cause keratoconjunctivitis sicca (CCS), a common degenerative eye disease that directly affects the quality of life of dogs and men, with no cure to date. This way, it is necessary research therapies that lead to permanent cure. The objective of this research is to analyze the efficacy of the topical use of mesenchymal stem cells (C-TM) heterologous derived from adipose tissue in the treatment of canine CCS in dogs. 40 dogs, divided into two groups will be used. Control group (CG, n = 20) comprised of 20 healthy dogs without ocular disease to obtain normal parameters. The treatment group (TG; n = 20) will include dogs with bilateral positive diagnosis of SCC with topical treatment of C-TM. The animals will be evaluated by the Lacrimal Schirmer Test, Fluorescein Test, Rose Bengal Test, Break Test of Lachrymal Film, Cytology Exfoliative of Conjunctiva, Cytology Aspirative with Fine Needle of the third eyelid gland and conjunctival tissue biopsy. The GT animals are topically treated with 50µl (50x106 C-TM) in both eyes, 4 applications with intervals of 7 days, and lubricant drops propylene glycol-based, 1 drop three times daily to both eyes for 30 days. The GT animals will be followed for a period of 180 days. The parametric data will be compared between movements and groups by analysis of variance (ANOVA) by using the Tukey method. In the case of non-parametric data will be used the Friedman and Kruskal-Wallis Tests to compare between moments and groups, respectively. The level of significance will be p <0.05. (AU)
