Abstract

Studies have demonstrated that the average life expectancy of patients with rheumatoid arthritis (RA) is lower than in healthy persons because of increased morbi-mortality due cardiovascular diseases. Possibly, this increased morbi-mortality occurs in consequence of the RA-related intense inflammatory process, that may result in endothelial dysfunction. Evidences also have suggested the involvement of the renin-angiotensin system in the cardiovascular damage related not only to the RA but also in the experimental models that simulate RA. It is believed that the angiotensin II (Ang II) actions on AT1 receptors may deepen the arthritis-related endothelial dysfunction. In parallel, the literature also reports the influence of androgens, especially testosterone, on the actions of Ang II on vascular tissues. Thus, this project aims to verify whether drastic reductions in serum testosterone levels may influence the pathophysiological actions of AIA in rat aortas. In order to meet this objective, the expression of AT1 and AT2 receptors as well as of the enzymes that are regulated by these receptors (NOX2, NOX4, iNOS and eNOS) will be investigated in aortas taken from control and AIA animals, either SHAM or orquidectimized. Additionally, it will be investigated relations between changes in the expression of AT1 and AT2 receptors as well as of the enzymes that are regulated by these receptors (NOX2, NOX4, iNOS and eNOS) and changes in responses of the studied aortas to Ang II and ACh.
