Abstract

Introduction: Ischemia and reperfusion phenomenon occurred in free flaps transfers. This phenomenon could cause flap necrosis. In diabetic condition this risk increases more. The aim of this project is the evaluation of ADSC effect on the viability of abdominal flap submitted to ischemia in diabetic rats.Material and Methods: ADSC's were isolated from isogenic Kioto rats and cultured in vitro. 18 rats were induced to diabetes (streptozotocin 50mg/kg, iv ) for eight weeks. After this period all animals were divided in three groups: group I - abdominal flap without ischemia; group II- abdominal flap with ischemia (3h) and buffered solution injection, abdominal flap with ischemia and buffered solution plus ADSC's injection. After seven days flap survival was assessed. Microvessel density was evaluated by hematoxilin eosin stain and semi quantitative real time polymerase chain reaction addressed gen expression of VEGF-a, iNOs, ECAM-1, C5ar. (AU)
