Abstract

Escherichia coli genome encodes seven different sigma factors. The primary sigma factor, Ã70 , is responsible for the transcription of most genes, in particular those associated with bacterial growth. RpoS (ÃS ) is the second in importance, being involved in the transcription of genes associated with the general stress response. Despite its relevance, the rpoS gene is very polymorphic and protein levels of this sigma factor are very variable. Moreover, the frequency of null mutations in rpoS is much higher than expected for spontaneous mutations. This is because there is a constant strong selective pressure on rpoS and on other genes that govern its expression. In this project, we will evaluate the level of rpoS polymorphism in the species E. coli in a longitudinal study of 12 months. Monthly samples will be analyzed for the presence of RpoS and for its intrinsic level, genetic polymorphism, frequency of spontaneous and adaptive mutations, fitness and other characteristics. As a control, a metagenomics analysis of rpoS will be performed. These data will be correlated with the rpoS genotype and RpoS level of the isolates. In parallel, the mechanism by which RpoS inhibits the transcription of Ã70 dependent genes will be investigated. (AU)
