Vegetative structure of the achlorophyllous orchid Pogoniopsis schenckii (Vanilloideae) and its interaction with mycorrhizal fungi

Abstract

Pogoniopsis, a little-known genus of the Orchidaceae, consists of two achlorophyllous species, with obligatory myco-heterotrophic metabolisms, it has a pale yellow color and develop in the organic matter in dense tropical forests. Myco-heterotrophic (MH) plants depend entirely on the association with mycorrhizal fungi throughout their lives cycle. This association is required for obtaining carbohydrates and other nutrients such as N and P, for example. The phylogenetic relationships of Pogoniopsis remain uncertain to date and studies of this MH species may be important for elucidating the evolutionary history of Vanilloideae and it can also contribute to gain understanding the evolution and specialization of these plants. The objectives of this project are to understand the organization of the underground vegetative structure and observe the pattern of fungal colonization associated with this kind MH through analysis in optical microscopy and scanning and transmission electron microscopy. The fungi present in these subterranean orchid organs will be isolated and identified. In addition, it will be assessed whether the isolated fungi will be capable of forming mycorrhizal association with the embryo in the mature seed and promote protocorms development. In addition, it will be analyzed the isotopic composition of the plant biomass, the carbon (´13C) and nitrogen (´15N) ratios, in order to assist the interpretation of the MH nature of this species indicating a possible specificity to certain fungal hosts and the origin of the ultimate C and N sources for this rare orchid species.
