Comparison of the effect of low-level laser therapy and diclofenac on inflammatory process, tissue repair and biochemical aspects and morphological aspects in an experimental model of skeletal muscle trauma in diabetic rats

Abstract

Diabetes mellitus is a heterogeneous group of metabolic disorders that have in common hyperglycemia, characterized by inadequate insulin secretory response, defects in insulin action, or both. This situation may result in delays in the repair process after skeletal muscle injury by changing morphology, resulting in decreased strength and may cause functional loss in varying degrees. Different types of therapies, pharmacological and non-pharmacological approaches have been used in the treatment and recovery of muscle injuries, such as anti-inflammatory drugs and low-level laser therapy. However the process of functional recovery and biochemical responses after an injury in the course of diabetes remains unknown. The aim of this work is to verify the inflammatory process, tissue repair and functional aspects of muscle injury resulting from trauma induced in rats with diabetes. A trauma will performed, on the right pelvic limb. Induction of diabetes is by administration of streptozotocin (50 mg / kg). Irradiation will be held at area of trauma (3J), one hour after injury protocol, per 30s. Diclofenac will be applied one hour after injury protocol (11.6 mg g -1) topically. Functional assessment, histological and morfometric analysis will be performed at 12, 24, 48, 72 and 96h and 14 and 28 days after trauma. Analysis of gene expression (TNF-±, IL-1², IL-6, IL-10 and miostatin) will be quantification by real-time polymerase chain reaction (RT-PCR), on skeletalmuscle at 12, 24, 48, 72 and 96h and 14 and 28 days after injury protocol.
