Use of detomidine, xylazine and oxytocin for pharmacologically-induced ex copula ejaculation in stallions

Abstract

Currently, the most widely used biotech is the collection of artificial vagina, however, as this method mimics the natural breeding, several physical, reproductive and / or behavioral preclude this method of collection. Thus, without triggering the ejaculation process it is not possible to obtain the semen, making it impossible to use these animals for breeding. Triggering neurophysiological mechanisms of ejaculation are still not completely understood, but it is already known that the activation of the receptor agonists ±1 and ±2 adrenergic, through the autonomic nervous system, is directly related to the ejaculatory process., It is also believed that the hormones estrogen and oxytocin actively participate in the ejaculation process by inducing rhythmical contractions in epididymis and seminiferous tubules. Studies previously developed using imipramine, and xylazine association protocols, showed high variability of results and low success rates in the induction of ejaculation. The aim of the study is to pharmacologically-induced ex copula ejaculation in stallions through the association of detomidine, xylazine and oxytocin. For this, 20 male horses will be used, not neutered, up 3 years old, previously collected by artificial vagina. Then, four protocols will be tested: P1 group - detomidine (4 mcg / kg); P2 group - xylazine (0.3 mg / kg) P3 group - detomidine (4 mcg / kg) + xylazine (0.3 mg / kg), group P4 - detomidine (4 mcg / kg) + xylazine (0.3 mg / kg) + oxytocin (20 IU). After collection, the semen will be evaluated: volume, color, pH, osmolarity and sperm concentration, sperm kinetics (HTR-IVOS-10 - Hamilton Thorn Research - Animal Version 12.0 L, USA) and morphofunctional evaluation by flow cytometry. For statistical analysis of the data will be used analysis of variance (ANOVA), the GLM procedure of SAS statistical program (2001), considering the fixed effects of treatment and evaluation moments. The averages are compared by Tukey test at the 5% level of significance. (AU)
