Abstract

The aim of this study is standardization the molecular diagnosis to determine human malaria parasites infectivity in different species of anopheline. For the standardization of the PCR, we will use anopheline ELISA positive results for the repetitive region of the circunsporozoite protein (CSP) of each kind of human malaria parasite, and each variant of P. vivax (VK210, VK247 and P. vivax-like), storage on the Evandro Chagas Institute, Belém, Pará The sequences of the CSP are available in the GenBank database (VK210 - M11926; VK247 - M69061; P. vivax-like - L13724, P. falciparum - DQ521732; P. malariae - J03992). The specificity is achieved using the BLAST program (http://www.ncbi.nlm.nih.gov/BLAST/). A conformational analysis will be performed to investigate the possibility of training for secondary structures. All oligonucleotídeos initiators are synthesized by RTD - Integrated DNA Technologies (USA). The enzymes will be selected containing at least one site of the cleavage products of amplification, using the NEBcutter V 2.0 program (http://tools.neb.com/NEBcutter2/index.php), resulting in fragments of DNA easily visible in the polyacrylamide gel. The use of this technique will be important for the evaluation of the anophelines`s infectivity, contributing to a better understanding of the factors malaria`s epidemiology, allowing the development of new strategies to control the disease.
